PYROSTAR is Fujifilm Wako’s range of endotoxin testing instruments and reagents, based on the Limulus Amoebocyte Lysate (LAL) assay.
The LAL assay operates using lysate isolated from the blood of the Atlantic Horseshoe Crab (Limulus polyphemus). When exposed to bacterial endotoxins, generally arising from lipopolysaccharides (LPS) in the bacterial cell wall, these lysates coagulate and in turn cause the crab’s blood to clot. The Pyrostar LAL assay harnesses this process to provide the most sensitive endotoxin detection method.
This PYROSTAR KY Single Test Colorimetric Endotoxin Test Kit provides 25 single test vials, each allowing for a single test using the Kinetic Colorimetric Assay (KCA) method with a sensitivity of 0.0002EU/mL, the most sensitive of the PYROSTAR assays. β-1,3-glucan has been shown to interfere with the reaction at very low levels, though this effect is inhibited in larger amounts. To avoid this potential for cross-reactivity KY contains large amounts of β-1,3-glucan, removing the possibility of inhibition.
Kinetic Colorimetric Assay (KCA) Method
The KCA method works by triggering a cascade of serine protease precursors on the detection of endotoxin. The end stage of this cascade is the hydrolysis of a chromogenic substrate by a clotting enzyme, resulting in the production of a yellow chromogen. Using a tube reader, such as the ET-7000 Toxinometer, or a microplate reader, the absorbance of the sample can be monitored and the time taken to reach a threshold, Ta, can be measured. From this the endotoxin concentration can be calculated.
