PYROSTAR is Fujifilm Wako’s range of endotoxin testing instruments and reagents, based on the Limulus Amoebocyte Lysate (LAL) assay.
The LAL assay operates using lysate isolated from the blood of the Atlantic Horseshoe Crab (Limulus polyphemus). When exposed to bacterial endotoxins, generally arising from lipopolysaccharides (LPS) in the bacterial cell wall, these lysates coagulate and in turn cause the crab’s blood to clot. The Pyrostar LAL assay harnesses this process to provide the most sensitive endotoxin detection method.
Historically endotoxin extraction has been carried out using water or saline, though the effectiveness of this has been questioned. FujiFilm Wako have improved the reliability of this method with their Endotoxin Extracting Solution. The addition of human serum albumin (HAS) improves the extraction and recovery of endotoxins, compared to previous water and saline methods. The solution is particularly recommended when testing samples that may contact blood or proteins. Overall this gives you more reliable, more accurate representative results that you can trust.
The solution can be used with the Gel Clot Assay (GCA), Kinetic Turbidimetric Assay (KTA) and Kinetic Colorimetric Assay (KCA) methods.
